NM_000240.4(MAOA):c.424G>T (p.Ala142Ser) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the MAOA gene (transcript NM_000240.4) at coding-DNA position 424, where G is replaced by T; at the protein level this means replaces alanine at residue 142 with serine — a missense variant. Submitter rationale: The A142S variant in the MAOA gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The A142S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In addition, this substitution occurs at a position that is conserved across species. However, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. Therefore, we interpret A142S as a variant of uncertain significance.
