This variant lies in the CACNA1S gene (transcript NM_000069.3) at coding-DNA position 1256, where G is replaced by A; at the protein level this means replaces arginine at residue 419 with histidine — a missense variant. Submitter rationale: The R419H variant in the CACNA1S gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R419H variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R419H as a variant of uncertain significance.
