This variant lies in the POMT2 gene (transcript NM_013382.7) at coding-DNA position 134, where C is replaced by G; at the protein level this means replaces proline at residue 45 with arginine — a missense variant. Submitter rationale: This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 45 of the POMT2 protein (p.Pro45Arg). This variant is present in population databases (rs753037500, gnomAD 0.06%). This variant has not been reported in the literature in individuals affected with POMT2-related conditions. ClinVar contains an entry for this variant (Variation ID: 451230). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated.
