This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 4274, where G is replaced by A; at the protein level this means replaces arginine at residue 1425 with lysine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the DSP gene. The R1425K variant has been reported in one patient with anthracycline-associated cardiomyopathy (AACM) (Wasielewski et al., 2014). The R1425K variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). However, the R1425K variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is only conserved in mammals and where lysine (K) is present as the wild type in multiple species. In silico analysis suggests that this variant likely does not alter the protein structure/function.
