Uncertain significance — the classification assigned by GeneDx to NM_000523.4(HOXD13):c.912C>G (p.Asp304Glu), citing GeneDx Variant Classification (06012015). This variant lies in the HOXD13 gene (transcript NM_000523.4) at coding-DNA position 912, where C is replaced by G; at the protein level this means replaces aspartic acid at residue 304 with glutamic acid — a missense variant. Submitter rationale: The D304E variant in the HOXD13 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The D304E variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The D304E variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret D304E as a variant of uncertain significance.
