Pathogenic — the classification assigned by GeneDx to NM_152296.5(ATP1A3):c.2806C>T (p.Gln936Ter), citing GeneDx Variant Classification (06012015): The Q936X variant in the ATP1A3 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The ATP1A3 variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret ATP1A3 as a pathogenic variant.
