NM_002047.4(GARS1):c.163A>G (p.Met55Val) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the GARS1 gene (transcript NM_002047.4) at coding-DNA position 163, where A is replaced by G; at the protein level this means replaces methionine at residue 55 with valine — a missense variant. Submitter rationale: The M55V variant in the GARS gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The M55V variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position that is conserved across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. Based on currently available evidence, we interpret M55V as a variant of uncertain significance.
