NM_021942.6(TRAPPC11):c.2168dup (p.Lys724fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.2168dupT variant in the TRAPPC11 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.2168dupT variant causes a frameshift starting with codon Lysine 724, changes this amino acid to a Glutamine residue, and creates a premature Stop codon at position 5 of the new reading frame, denoted p.Lys724GlnfsX5. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The c.2168dupT variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.2168dupT as a pathogenic variant.
