Submitter rationale: The c.2841_2843delCCT variant in the KIF11 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The c.2841_2843delCCT variant causes an in-frame deletion of one amino acid, Leucine 948, denoted p.Leu948del. This amino acid deletion occurs at a position that is conserved in mammals. In silico analysis predicts this variant is probably damaging to the protein structure/function. The c.2841_2843delCCT variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.2841_2843delCCT as a likely pathogenic variant.
