This variant lies in the KDM5B gene (transcript NM_006618.5) at coding-DNA position 340, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 114 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The Q114X variant in the KDM5B gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q114X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Based on currently available evidence, we interpret Q114X as a variant of uncertain significance.
