Uncertain significance — the classification assigned by GeneDx to NM_002968.3(SALL1):c.3601G>A (p.Gly1201Ser), citing GeneDx Variant Classification (06012015). This variant lies in the SALL1 gene (transcript NM_002968.3) at coding-DNA position 3601, where G is replaced by A; at the protein level this means replaces glycine at residue 1201 with serine — a missense variant. Submitter rationale: The G1201S variant in the SALL1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The G1201S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret G1201S as a variant of uncertain significance.
