This variant lies in the HUWE1 gene (transcript NM_031407.7) at coding-DNA position 2929, where A is replaced by G; at the protein level this means replaces lysine at residue 977 with glutamic acid — a missense variant. Submitter rationale: The K977E variant in the HUWE1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The K977E variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In addition, this substitution occurs at a position that is conserved across species. However, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret K977E as a variant of uncertain significance.
