Uncertain significance — the classification assigned by GeneDx to NM_006517.5(SLC16A2):c.811T>C (p.Phe271Leu), citing GeneDx Variant Classification (06012015): he Y345H variant in the SLC16A2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The Y345H variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals. In silico analysis predicts this variant likely does not alter the protein structure/function. We interpret Y345H as a variant of uncertain significance.
