This variant lies in the SIX1 gene (transcript NM_005982.4) at coding-DNA position 510, where C is replaced by A; at the protein level this means replaces asparagine at residue 170 with lysine — a missense variant. Submitter rationale: The N170K variant in the SIX1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The N170K variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The N170K variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret N170K as a variant of uncertain significance.
