This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 2560, where G is replaced by A; at the protein level this means replaces glycine at residue 854 with serine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the DSP gene. The G854S variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The G854S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is only conserved in mammals and where serine (S) is the wild type in multiple non-mammalian species. Finally, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
