Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_020297.4(ABCC9):c.2557G>A (p.Glu853Lys), citing Ambry Variant Classification Scheme 2023: The p.E853K variant (also known as c.2557G>A), located in coding exon 21 of the ABCC9 gene, results from a G to A substitution at nucleotide position 2557. The glutamic acid at codon 853 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
