This variant lies in the SCN2A gene (transcript NM_001040142.2) at coding-DNA position 5465, where C is replaced by T; at the protein level this means replaces alanine at residue 1822 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 1822 of the SCN2A protein (p.Ala1822Val). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and valine. This variant is present in population databases (rs757542048, ExAC 0.009%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
