Uncertain significance — the classification assigned by GeneDx to NM_004415.4(DSP):c.4623C>G (p.Asp1541Glu), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the DSP gene. The D1541E variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). However, the D1541E variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to aspartic acid are tolerated across species and where glutamic acid is present as the wild type in several species. Finally, in silico analysis suggests that this variant likely does not alter the protein structure/function.
