This variant lies in the TOPORS gene (transcript NM_005802.5) at coding-DNA position 74, where C is replaced by T; at the protein level this means replaces serine at residue 25 with leucine — a missense variant. Submitter rationale: The S25L variant in the TOPORS gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S25L variant is observed in 5/16314 (0.03%) alleles from individuals of South Asian background in the ExAC dataset (Lek et al., 2016). The S25L variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret S25L as a variant of uncertain significance.
