Uncertain significance — the classification assigned by GeneDx to NM_020778.5(ALPK3):c.1214CAGGCC[3] (p.403PG[4]), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the ALPK3 gene. The c.1826_1831dupCAGGCC variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The c.1826_1831dupCAGGCC variant results in an in-frame duplication of two amino acids. However, this region is not conserved and in silico analysis predicts the duplication of these residues likely does not alter the protein structure/function.
