NM_003105.6(SORL1):c.2401T>C (p.Cys801Arg) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the SORL1 gene (transcript NM_003105.6) at coding-DNA position 2401, where T is replaced by C; at the protein level this means replaces cysteine at residue 801 with arginine — a missense variant. Submitter rationale: The C801R variant in the SORL1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The C801R variant is observed in 1/66636 (0.0015%) alleles from individuals of non-Finnish European background in the ExAC dataset (Lek et al., 2016). The C801R variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret C801R as a variant of uncertain significance.
