This variant lies in the PDZD7 gene (transcript NM_001195263.2) at coding-DNA position 244, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 82 with asparagine — a missense variant. Submitter rationale: The D82N variant in the PDZD7 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is observed in 17/10334 (0.16%) alleles from individuals of African background in the ExAC dataset (Lek et al., 2016). The D82N variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position where amino acids with similar properties to Aspartic Acid are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret D82N as a variant of uncertain significance.
