NM_020247.5(COQ8A):c.397C>T (p.Gln133Ter) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The Q133X nonsense variant in the ADCK3 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q133X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Although this variant has not been reported previously to our knowledge, it is interpreted to be a likely pathogenic variant.
