Pathogenic — the classification assigned by GeneDx to NM_001368894.2(PAX6):c.844G>T (p.Glu282Ter), citing GeneDx Variant Classification (06012015): The E268X nonsense variant in the PAX6 gene has been reported previously in association with aniridia (Yokoi et al., 2016). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016). In summary, we consider this variant to be pathogenic.
