NM_020335.3(VANGL2):c.971G>A (p.Arg324Gln) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The R324Q variant in the VANGL2 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R324Q variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R324Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R324Q as a variant of uncertain significance.
