Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001256789.3(CACNA1F):c.868C>T (p.Arg290Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 290 of the CACNA1F protein (p.Arg290Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with Cone-rod dystrophy, night blindness (PMID: 28341476, 32531858, 37206923). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt CACNA1F protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change affects CACNA1F function (PMID: 37206923). For these reasons, this variant has been classified as Pathogenic.
