Pathogenic — the classification assigned by GeneDx to NM_000397.4(CYBB):c.210dup (p.Val71fs), citing GeneDx Variant Classification (06012015): The c.210dupA variant in the CYBB gene has been reported previously in association with X-linked chronic granulomatous disease (Roos et al., 2010). The duplication causes a frameshift starting with codon Valine 71, changes this amino acid to a Serine residue and creates a premature Stop codon at position 32 of the new reading frame, denoted p.Val71SerfsX32. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). In summary, we consider this variant to be pathogenic.
