The variant allele was found at a frequency of 0.00058 in 251116 control chromosomes, predominantly at a frequency of 0.00076 within the Non-Finnish European subpopulation in the gnomAD database, including 1 homozygote. The observed variant frequency within Non-Finnish European control individuals in the gnomAD database exceeds the estimated maximal expected allele frequency for disease-causing variants in TRAPPC11. c.2147C>G has been reported in the literature in an individual suspected of a neuromuscular disease without a conclusive diagnosis (Barbosa-Gouveia_2022) or in a compound heterozygous individual carrying second variant classified as VUS in ClinVar (e.g. Colona_2025). These reports do not provide unequivocal conclusions about association of the variant with Limb-Girdle Muscular Dystrophy, Autosomal Recessive. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
