NM_000525.4(KCNJ11):c.980C>A (p.Ser327Tyr) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): The S327Y variant in the KCNJ11 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S327Y variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is not conserved. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret S327Y as a variant of uncertain significance.
