Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant allele was found at a frequency of 2.4e-05 in 246386 control chromosomes. c.5087C>T has been reported in the literature in at-least two compound heterozygous individuals affected with Short-rib thoracic dysplasia (example: Zhang_2018, Stals_2018). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
