Likely pathogenic for Gastrointestinal stromal tumor — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NM_003000.3(SDHB):c.575G>C (p.Cys192Ser), citing LMM Criteria: The Cys192Ser variant in SDHB has not been previously reported in any other fami lies with hereditary paragangliomas and pheochromocytomas or GIST. This variant has not been identified in large population studies. Computational analyses (bio chemical amino acid properties, conservation, AlignGVGD, PolyPhen2, and SIFT) su ggest that this variant may impact the protein, though this information is not p redictive enough to determine pathogenicity. Finally, similar variants at the sa me position (Cys192Arg, Cys192Tyr) have been reported in individuals with heredi tary paragangliomas and pheochromocytomas (Neuman 2002, Benn 2006, Lefebre 2012) . While collectively, this information supports that the Cys192Ser variant is li kely pathogenic, additional studies are needed to fully establish its clinical s ignificance.
