Likely pathogenic for Torsion dystonia 6 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_018105.3(THAP1):c.238A>G (p.Ile80Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 80 of the THAP1 protein (p.Ile80Val). This variant is present in population databases (rs372080941, gnomAD 0.02%). This missense change has been observed in individuals with dystonia (PMID: 21847143, 22377579, 26087139). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 444751). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
