NM_018105.3(THAP1):c.270_273del was classified as Pathogenic for Torsion dystonia 6 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change results in a premature translational stop signal in the THAP1 gene (p.Glu91Ilefs*28). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 91 amino acids of the THAP1 protein. This variant is present in population databases (rs749414480, ExAC 0.002%). This variant has not been reported in the literature in individuals with THAP1-related conditions. This variant disrupts the C-terminus of the THAP1 protein. Other variant(s) that disrupt this region (p.Arg169*) have been determined to be pathogenic (Invitae). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. For these reasons, this variant has been classified as Pathogenic.
