Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001061.7(TBXAS1):c.263T>C (p.Ile88Thr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: TBXAS1 c.263T>C (p.Ile88Thr) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 0.00033 in 251424 control chromosomes in the gnomAD database, including 1 homozygotes. c.266T>C has been reported in the literature in individuals affected with Ghosal Hematodiaphyseal Dysplasia (Sharma_2018). This report does not provide unequivocal conclusions about association of the variant with Ghosal Hematodiaphyseal Dysplasia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication have been ascertained in the context of this evaluation (PMID: 28868793).Three submitters have cited clinical-significance assessments for this variant to ClinVar after 2014.
