This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 67, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 23 with glutamine — a missense variant. Submitter rationale: This missense variant replaces glutamic acid with glutamine at codon 23 of the BRCA1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). This variant has been reported to be functional in a haploid cell proliferation assay (PMID: 30209399). This variant has been reported in individuals and families affected with breast and/or ovarian cancer (PMID: 21918854, 32380732, 32856854). This variant has been identified in 1/250924 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
