NM_000059.4(BRCA2):c.7552C>G (p.Leu2518Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This missense variant replaces leucine with valine at codon 2518 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function. Functional studies have reported that this variant does not impact BRCA2 function in cell proliferation assays (PMID: 35190686, 39779857) and in sensitivity assays to cisplatin and PARP inhibitor (PMID: 39779848). To our knowledge, this variant has not been reported in individuals affected with BRCA2-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
