This variant lies in the BRCA1 gene (transcript NM_007294.4) at coding-DNA position 2586, where G is replaced by C; at the protein level this means replaces lysine at residue 862 with asparagine — a missense variant. Submitter rationale: The p.K862N variant (also known as c.2586G>C), located in coding exon 9 of the BRCA1 gene, results from a G to C substitution at nucleotide position 2586. The lysine at codon 862 is replaced by asparagine, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6502 samples (13004 alleles) with coverage at this position. To date, this alteration has been detected with an allele frequency of approximately 0.0004% (greater than 225000 alleles tested) in our clinical cohort. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of p.K862N remains unclear.
