NM_000527.5(LDLR):c.1046dup (p.Arg350fs) was classified as Pathogenic for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.1046dupA pathogenic mutation, located in coding exon 7 of the LDLR gene, results from a duplication of A at nucleotide position 1046, causing a translational frameshift with a predicted alternate stop codon (p.R350Afs*8). This alteration has been observed in an individual with a personal history consistent with familial hypercholesterolemia (Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
