Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.10768G>A (p.Glu3590Lys), citing Ambry Variant Classification Scheme 2023: The p.E3590K variant (also known as c.10768G>A), located in coding exon 26 of the APOB gene, results from a G to A substitution at nucleotide position 10768. The glutamic acid at codon 3590 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
