NM_018979.4(WNK1):c.2401A>G (p.Ile801Val) was classified as Uncertain significance for Neuropathy, hereditary sensory and autonomic, type 2A; Pseudohypoaldosteronism type 2C by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The valine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with WNK1-related disease. ClinVar contains an entry for this variant (Variation ID: 440420). This variant is present in population databases (rs139449198, ExAC 0.2%). This sequence change replaces isoleucine with valine at codon 801 of the WNK1 protein (p.Ile801Val). The isoleucine residue is moderately conserved and there is a small physicochemical difference between isoleucine and valine.
