Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Thr18 amino acid residue in GJB1. Another variant that disrupts this residue have been observed in affected individuals (Invitae), suggesting that it is a clinically significant residue. As a result, variants that disrupt this residue are likely to be causative of disease. This sequence change replaces threonine with alanine at codon 18 of the GJB1 protein (p.Thr18Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with GJB1-related disease. ClinVar contains an entry for this variant (Variation ID: 439756).
