This variant lies in the FBN1 gene (transcript NM_000138.5) at coding-DNA position 1175, where C is replaced by T; at the protein level this means replaces proline at residue 392 with leucine — a missense variant. Submitter rationale: This missense variant replaces proline with leucine at codon 392 of the FBN1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual suspected of having Marfan syndrome and has been shown not to segregate with disease in this family (PMID: 30032985). This variant has been identified in 1/250242 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
