Likely benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001134363.3(RBM20):c.1027C>T (p.His343Tyr), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the RBM20 gene (transcript NM_001134363.3) at coding-DNA position 1027, where C is replaced by T; at the protein level this means replaces histidine at residue 343 with tyrosine — a missense variant. Submitter rationale: Variant summary: RBM20 c.1027C>T (p.His343Tyr) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00012 in 154822 control chromosomes. The observed variant frequency is approximately 3 fold of the estimated maximal expected allele frequency for a pathogenic variant in RBM20 causing Dilated Cardiomyopathy phenotype (4.7e-05). To our knowledge, no occurrence of c.1027C>T in individuals affected with Dilated Cardiomyopathy and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 43963). Based on the evidence outlined above, the variant was classified as likely benign.
