NM_006231.4(POLE):c.1019A>T (p.Glu340Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E340V variant (also known as c.1019A>T), located in coding exon 10 of the POLE gene, results from an A to T substitution at nucleotide position 1019. The glutamic acid at codon 340 is replaced by valine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
