Pathogenic — the classification assigned by GeneDx to NM_003506.4(FZD6):c.346C>T (p.Arg116Ter), citing GeneDx Variant Classification (06012015): The R116X variant in the FZD6 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The R116X variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016). We interpret R116X as a pathogenic variant.
