Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022455.5(NSD1):c.5927T>C (p.Ile1976Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 1976 of the NSD1 protein (p.Ile1976Thr). For these reasons, this variant has been classified as Pathogenic. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt NSD1 protein function. ClinVar contains an entry for this variant (Variation ID: 436064). This missense change has been observed in individual(s) with Sotos syndrome and features consistent with an NSD1-related condition (PMID: 22924495; Invitae). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency).
