Submitter rationale: This sequence change replaces arginine, which is basic and polar, with lysine, which is basic and polar, at codon 1082 of the KANK1 protein (p.Arg1082Lys). This variant also falls at the last nucleotide of exon 6, which is part of the consensus splice site for this exon. This variant is present in population databases (rs201207109, gnomAD 0.07%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with KANK1-related conditions.
