NM_000426.4(LAMA2):c.5675A>T (p.Gln1892Leu) was classified as Uncertain significance for LAMA2-related muscular dystrophy by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with leucine at codon 1892 of the LAMA2 protein (p.Gln1892Leu). The glutamine residue is moderately conserved and there is a moderate physicochemical difference between glutamine and leucine. This variant is present in population databases (rs779542581, ExAC 0.002%). This variant has not been reported in the literature in individuals with LAMA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 435707). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
