This variant lies in the AHI1 gene (transcript NM_001134831.2) at coding-DNA position 983, where G is replaced by T; at the protein level this means replaces serine at residue 328 with isoleucine — a missense variant. Submitter rationale: The S328I variant in the AHI1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The S328I variant is observed in 26/30780 (0.084%) alleles from individuals of South Asian background, in large population cohorts (Lek et al., 2016). The S328I variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret S328I as a variant of uncertain significance.
