NM_001378454.1(ALMS1):c.7526G>A (p.Arg2509Gln) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R2510Q variant (also known as c.7529G>A), located in coding exon 8 of the ALMS1 gene, results from a G to A substitution at nucleotide position 7529. The arginine at codon 2510 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
